Tirzepatide is a synthetic 39-residue lipidated peptide based on the GIP sequence that acts as an agonist at the GIP and GLP-1 receptors. It has been studied in preclinical models and in large clinical trial programmes (SURPASS, SURMOUNT).
Reported as a dual GIP and GLP-1 receptor agonist; in mice it activated both receptors in vitro and produced glucose-dependent insulin secretion, with greater effects on body weight and food intake than a GLP-1 receptor agonist (Coskun 2018).
Approved in the US (FDA) as Mounjaro (type 2 diabetes) and Zepbound (chronic weight management; obstructive sleep apnea in adults with obesity) per current DailyMed labels.
